Traceability

The balances used for gravimetric measurements are calibrated with weights traceable to the national standards (DKD). The calibration of the balances is verified daily internally and annually by an external accredited calibration service. Homogeneity

Random replicate samples of the final packaged RM have been analysed to prove homogeneity compliant with ISO 17034.

Storage

The RM should be stored in the original sealed container at the indicated temperature.

Instructions for use

It is recommended to use 1 mg as the minimum sample size and if less material is used, to increase the certified uncertainty by a factor of two for half sample and four for a quarter of sample. If storage after opening is necessary, the RM should be tightly closed and kept from light and moisture. If the RM was in a sealed ampoule, it should be transferred to a vial with minimum head space.

核心檢測方法
采用堿性過硫酸鉀消解結(jié)合紫外分光光度法。消解后的樣品在220nm和275nm波長處測定吸光度,通過標(biāo)準(zhǔn)曲線法計算總氮含量。
檢出限與定量限
方法檢出限為0.05mg/L,定量限為0.20mg/L。當(dāng)使用四氯化碳萃取時,需驗證回收率是否滿足70%-120%的要求。
質(zhì)控樣品要求
每批次樣品應(yīng)包含至少10%的平行樣、空白樣和加標(biāo)回收樣。四氯化碳純度需≥99.9%,并定期進(jìn)行空白實驗以驗證試劑干擾。
關(guān)鍵實驗步驟
1. 樣品保存:水樣需用硫酸酸化至pH≤2,4℃冷藏并7日內(nèi)完成分析;
2. 消解程序:在120℃條件下消解30分鐘,冷卻后加入四氯化碳萃取干擾物質(zhì);
3. 比色測定:使用10mm石英比色皿,扣除275nm處的背景干擾。
特別說明
1. 四氯化碳屬于有毒試劑,操作需在通風(fēng)櫥中進(jìn)行;
2. 當(dāng)水樣含有大量油脂或色度時,需增加四氯化碳萃取次數(shù);
3. 標(biāo)準(zhǔn)物質(zhì)需使用國家認(rèn)證的氮標(biāo)準(zhǔn)溶液,臨用現(xiàn)配。
