培養(yǎng)基：EMEM完全培養(yǎng)基：90%EMEM+10%FBS
培養(yǎng)條件：培養(yǎng)溫度37℃,；氣體環(huán)境5%CO2+95%空氣；
培養(yǎng)方法：復蘇步驟：①凍存管從液氮或-80℃冰箱中取出放入PE手套中，迅速沒入37℃水浴鍋,，搖晃凍存管加速溶解,，以1min內全部溶解為宜；②在超凈臺中將溶解好的細胞液加入到裝有9mL完全培養(yǎng)基的離心管內,，1000-1200rpm離心5min,，棄去上清液，用1-2mL完全培養(yǎng)基重懸細胞,。③將細胞懸液加入到含有5-6mL完全培養(yǎng)基的T25瓶中,，放入培養(yǎng)箱培養(yǎng)。
細胞傳代：①將舊培養(yǎng)液吸除,，PBS清洗兩遍后,，加入1-2mL胰酶（0.25%Trypsin+0.02%EDTA）；②鏡下觀察消化情況,，在細胞邊緣縮小,，貼壁松動時（可用吸管吸起些許胰酶輕輕吹打細胞層某處，肉眼可見細胞層脫落,，即消化完成,，否則繼續(xù)消化），直接吸掉胰酶,，加入5-6mL完全培養(yǎng)基,，輕輕吹打細胞層，把細胞層吹落,，吹散,。③將細胞懸液按1:2比例分到新的T25瓶中，添加適當的完全培養(yǎng)基,，把細胞懸液打勻,，于培養(yǎng)箱中培養(yǎng)。,；④注意培養(yǎng)基pH值變化和細胞密度,，定期換液（每周2-3次），待細胞密度達到80%-90%時,，重復傳代操作或者凍存,。
