培養(yǎng)基:EMEM完全培養(yǎng)基:90%EMEM+10%FBS
培養(yǎng)條件:37℃,;5%CO2+95%空氣,;
培養(yǎng)方法:復蘇步驟:①凍存管從液氮或-80℃冰箱中取出放入PE手套中,迅速沒入37℃水浴鍋,搖晃凍存管加速溶解,,以1min內全部溶解為宜,;②在超凈臺中將溶解好的細胞液加入到裝有9mL完全培養(yǎng)基的離心管內,1000-1200rpm離心5min,,棄去上清液,,用1-2mL完全培養(yǎng)基重懸細胞。③將細胞懸液加入到含有5-6mL完全培養(yǎng)基的T25瓶中,,放入培養(yǎng)箱培養(yǎng),。
細胞傳代:①將舊培養(yǎng)液吸除,PBS清洗兩遍后,,加入1-2mL胰酶(0.25%Trypsin+0.02%EDTA),;②鏡下觀察消化情況,在細胞邊緣縮小,,貼壁松動時(可用吸管吸起些許胰酶輕輕吹打細胞層某處,,肉眼可見細胞層脫落,即消化完成,,否則繼續(xù)消化),,直接吸掉胰酶,加入5-6mL完全培養(yǎng)基,,輕輕吹打細胞層,,把細胞層吹落,吹散,。③將細胞懸液按1:2比例分到新的T25瓶中,,添加適當的完全培養(yǎng)基,把細胞懸液打勻,,于培養(yǎng)箱中培養(yǎng),。④注意培養(yǎng)基pH值變化和細胞密度,定期換液(每周2-3次),,待細胞密度達到80%-90%時,,重復傳代操作或者凍存。
